| preparation method | at 10 ℃, 10mmol of D-alanine is dissolved in 4.0ml of sodium hydroxide (NaOH) water soluble solution with mass concentration of 8% to obtain the dissolved solution, while stirring, 14mmol (I .e. 1.8ml) of benzyl chloroformate was added dropwise to dissolved solution at a rate of 1 drop/9 seconds for reaction, and 8% sodium hydroxide (NaOH) aqueous solution with a mass concentration was added dropwise to keep the PH at 8. after dropping, was placed at 28 ℃, stirred for 15 hours, thin layer detection was performed, and the solution was completely reacted, wash polyester with 5ml/time of ether for 3 times, discard the ether, and get the aqueous phase. adjust the PH of the aqueous phase to 3 with concentrated hydrochloric acid with mass concentration of 30%, and appear milky white solid. extract the liquid with 5ml/time of ethyl acetate for 3 times and merge, the combined liquid is washed twice with 5ml/time of distilled and once with 5ml of saturated salt water, then dried with anhydrous magnesium sulfate for 12 hours, filtered by suction, and the filtrate is concentrated under reduced pressure to obtain a light yellow oil. The mixed solvent prepared by ethyl acetate and petroleum ether with a volume ratio of 1:3 is used as the mobile phase, and purified by chromatography column, N-benzyloxycarbonyl-D-alanine, yield: 71.8%, melting point Mp:86-87 degrees C. |
